DAT Detects Bound Antibody On Patient Red Cells
- The DAT tests patient red cells directly for IgG and complement bound to the cells.
- It is extremely sensitive and can detect very small amounts of IgG on red cells.
Always Wash Cells Before Adding AHG
- Wash patient red cells before adding antihuman globulin to remove unbound antibody in plasma.
- Skip washing and free antibody can neutralize reagent and often cause false-negative DATs.
Use Poly-Specific AHG As A Screen
- Poly-specific AHG contains both anti-IgG and anti-C3 and is used as an initial screen.
- Mono-specific reagents (anti-IgG and anti-C3 separately) follow a positive poly-specific screen to define what is coating the cells.
