NOTE

Exploring Daily Variation and Methylation Impact on Klotho Levels

The daily variation in Klotho levels is not completely understood, with questions arising about whether it is influenced by consumption, turnover rate, or supply depletion. The regulation of daily levels remains uncertain, considering factors like degradation and expression. Although the half-life of Klotho is short in mice, it is longer in humans and non-human primates, suggesting a complex production and degradation process. The diurnal variation in Klotho levels is observed in aging, possibly linked to hypermethylation around its promoter region, which impairs transcription and translation. This suggests a need to explore methods to counteract the methylation process in order to preserve Klotho levels.
