The Number of the Beast
Revelation 13 is where we learn that John has his bullseye set on the Roman Empire. He uses an ancient Jewish form of numerology called Gematria to assign a numerical value to every letter in the alphabet. If you take Caesar Nero's name and transliterate it into Greek, then you would take this name and its value would come to 666 if you use this technique of Hebrew numerology. So it's widely regarded that this mark of the beast, this number of the beast is in fact referring to Nero.
