I'm not sure there are many plausible scenarios where something like a supervolcano, supernova, or asteroid could get every human. I would argue that at this point anything that reduces Earth to only microorganisms sets off a 5 million year timer. That's about how long it was since the first animal showed up to now. This is long before our sun goes red giant and extinguishes life that way.
