In the klonwar, season seven, darthmall seems to suspect that palpatin is grooming anikin as his new apprentice. So he probably figures it out when darth wader shows up, who's inside that suit? Tarkin figures it out too. Vader might very well be jeddi night, annikan skywalker whom tarkin had fought beside during the kloan wars and for whom he had developed a grudging appreciation.
