What's the Place Marker for Downtown?

In landlock cities, what institutes downtown? In my city, downtown is always around the harbor and is often the oldest part of town. How is there a downtown with no harbor? Please help i mean, there are some midwestern cities where i feel like the downtown was created more or less at random.
