A Protac Is a Proteolysis Targeting Chimera

A protac is a proteolysis targeting chimera it's a concept that came from the labs of cray crews at Yale and Ray Deshaes who was at Caltech as an academic before he became the head of research at Amgen. We envisioned a bifunctional molecule that on the one hand would bind to the extracellular target but then on the other hand it would bind a lysosomal trafficking shuttle. The first molecules we made that functioned as these lie tags were monoclonal antibodies that bind a target of interest chemically modified with manos 6 phosphate derivatives which we chemically synthesized so the antibody would bind the target  and the whole complex would
