Ibuprofen can block the bone formation response, which could be an important determinant of why bone sometimes fails to adapt as we might expect. We have done studies to show that NSAID use primarily before exercise could be deleterious for bone. Prostaglandin E2 is an essential signaling factor in the activation of bone formation.
