The next step after you get decent off-world production, which probably means lots of automation but doesn't have to, is simply patience. Everything about colonizing it galaxy once you get that first foothold off Earth is patience, patience, and more patience. I do not think we're going to see growth rates like we did in the 20th century, even if we did get radical life extension. If we did, by the year 2500, we would number a stunning thousand times more people than are alive now, 8 trillion. 500 times our current population.
