The body creates more and more immune cells with each shot of vaccine. That's why almost every immunization we get, like measles in a polio, takes more than one dose to be long lasting. Some months or even years after vaccination, these memory cells may be there kind of asleep, but with one click of an infection, they wake up and they explode into action.
