The prevailing biochemical methods, even dating back many decades for studying the fundamental packaging of chromatin into these nucleosomes are population averaged assays. And to ask questions at the level of individual fibers, you actually can't use an illuminous sequencer. So we'd like to understand how the fiber is structured. You can't address that question because the platform is fundamentally limited, right? Well let's look in a different direction and try PacBio sequencing.
