The Mechanism of Androgen Receptor Loss

The active compounds promote androgen receptor loss through a mechanism that depends on covalent modification of C145. But this effect is prevented rather than replicated by the genetic disruption of Nono. The cysteine belongs to an area nestled in between the two RNA binding domains of Nono, suggesting it's poised to have an impact on how Nono binds RNAs. And we were able to show if we mutate that cysteine to serine, the compound loses its activity.
